Sure, it's possible.

With bulldogs, you would need a DNA test to tell "who's who" (unless one is a pure red/rednose and one is a pure black/blacknose dominant), but it could be done.

Keep in mind, though, semen is antagonistic to other semen, so filling a bitch with two loads can cancel each other out at times ...
